Hrvatski znanstvenik Nenad Šestan predvodio je tim znanstvenika s prestižnog američkog Sveučilišta Yale koji su uspjeli oživjeti organe mrtve svinje, prenosi New York Times.

Eksperiment, čiji su rezultati zapanjili i znanstvenike koji nisu sudjelovali u njemu, odvijao se na sljedeći način. Svinje su ležale mrtve u laboratoriju sat vremena: nije bilo cirkulacije krvi, srca su prestala kucati te nije bilo nikakve moždane aktivnosti. Potom su znanstvenici s Yalea ubrizgali otopinu u tijela mrtvih svinja uz pomoć uređaja sličnog ECMO-u.

Iako svinje nisu bile ni na koji način u svjesnom stanju, njihove naizgled mrtve stanice su oživjele. Srca su im počela kucati dok je otopina, koju su znanstvenici nazvali OrganEx, cirkulirala njihovim venama i arterijama. Stanice u njihovim organima, uključujući srce, jetru, bubrege i mozak, opet su bile u funkciji, a životinje ni u jednom trenutku nisu bile ukočene, kao što bi to bio slučaj s pravom mrtvom svinjom.

Ostale svinje, koje su bile mrtve već sat vremena, priključili su na umjetna pluća (ECMO uređaj), aparat koji je pumpao krv u njihova tijela. Ukočile su se, organi su im natekli i pretrpjeli oštećenja, a krvne žile su kolabirale. Na leđima su imale ljubičaste mrlje uslijed nakupljanja krvi.

Cilj - organi za transplantaciju

Skupina znanstvenika rezultate eksperimentalnog istraživanja objavila je u časopisu Nature.

Tvrde da je cilj njihova istraživanja povećati zalihe organa za transplantaciju. Nadaju se da bi se njihova tehnologija mogla koristiti za sprečavanje teških oštećenja srca - nakon razornog srčanog udara - ili mozga nakon moždanog udara.

No, najnovija otkrića tek su prvi korak, a još je daleko mogućnost da bi se slična tehnologija mogla isprobati na ljudima, rekao je Stephen Latham, bioetičar sa Sveučilišta Yale, koji je blisko surađivao sa znanstvenicima.

Skupinu znanstvenika predvodio je ugledni hrvatski znanstvenik Nenad Šestan, profesor neuroznanosti, komparativne medicine, genetike i psihijatrije na Medicinskom fakultetu Yaleu

i on je bio zapanjen otkrićem. Posjetimo, prije tri godine, osvanuo je na naslovnicama vodećih svjetskih medija zahvaljujući sličnom, velikom otkriću. Šestan i njegovi suradnici uspjeli su uz pomoć visokotehnološkog sustava BrainEx, koji su sami razvili, obnoviti temeljnu staničnu aktivnost u svinjskom mozgu nekoliko sati nakon smrti životinje. Popularnim jezikom rečeno, uspjeli su "oživjeti" stanice mozga jednog sisavca četiri sata nakon smrti.

Jedan od autora studije je i dr. David Andrijević, mladi Osječanin, također neuroznanstvenik s Yalea, prenosi Jutarnji list. 

- Nismo znali što možemo očekivati. Bilo je nevjerojatno, rekao je.

Nova definicija smrti?

I drugi znanstvenici koji nisu sudjelovali u studiji bili su podjednako zadivljeni.

- Nevjerojatno, zapanjeni smo, rekla je Nita Farahany, profesorica prava na Dukeu koja poučava etičke, pravne i društvene implikacije novih tehnologija.

Dr. Farahany je rekla da rad postavlja pitanja o definiciji smrti.

Rad na projektu započeo je prije nekoliko godina kada je skupina napravila sličan eksperiment sa svinjskim mozgom.

Prema riječima još jednog člana tima iz Hrvatske, dr. Zvonimira Vrselje, otkriće iz 2019. ih je navelo da se zapitaju mogu li oživjeti cijelo tijelo.

Otopina OrganEx sadrži hranjive tvari, protuupalne lijekove, lijekove za sprečavanje smrti stanica, blokatore živaca - tvari koje prigušuju aktivnost neurona i sprečavaju svaku mogućnost da svinje dođu svijesti - te umjetni hemoglobin pomiješan s krvlju svake životinje.

Znanstvenici su poduzeli sve mjere opreza kako životinje ne bi patile. Svinje su dobile anesteziju prije nego što im je zaustavljen rad srca, a duboka anestezija je nastavljena tijekom postupka.

Osim toga, blokatori živaca u otopini OrganEx zaustavljaju oštećenje živaca kako bi se osiguralo da mozak ne bude aktivan. Znanstvenici su također hladili životinje kako bi usporili kemijske reakcije. Pojedinačne moždane stanice bile su žive, ali nije bilo naznaka bilo kakve organizirane živčane aktivnosti u mozgu.

Jedan detalj bio je osobito zapanjujući: svinje koje su liječene OrganExom trzale su glavama kada su istraživači ubrizgali kontrastno sredstvo za snimanje.

Dr. Latham naglašava da ne znaju zbog čega su se svinje trzale, jer nije bilo naznaka da je mozak potaknuo kretnje.

Yale je podnio zahtjev za patentiranjem nove tehnologije. Prema riječima Nenada Šestana, njihov cilj je vidjeti funkcioniraju li organi kako treba i mogu li se uspješno presaditi.

Kroz određeno vrijeme, znanstvenici se nadaju da će testirati može li tehnologija pridonijeti oporavku oštećenog srca ili mozga.

Nenad Šestan je zadarski znanstvenik koji radi na sveučilistu Yale u SAD-u

Hrvatski znanstvenik Nenad Šestan predvodio je tim znanstvenika s prestižnog američkog Sveučilišta Yale koji su uspjeli oživjeti organe mrtve svinje, prenosi New York Times.

Eksperiment, čiji su rezultati zapanjili i znanstvenike koji nisu sudjelovali u njemu, odvijao se na sljedeći način. Svinje su ležale mrtve u laboratoriju sat vremena: nije bilo cirkulacije krvi, srca su prestala kucati te nije bilo nikakve moždane aktivnosti. Potom su znanstvenici s Yalea ubrizgali otopinu u tijela mrtvih svinja uz pomoć uređaja sličnog ECMO-u.

Iako svinje nisu bile ni na koji način u svjesnom stanju, njihove naizgled mrtve stanice su oživjele. Srca su im počela kucati dok je otopina, koju su znanstvenici nazvali OrganEx, cirkulirala njihovim venama i arterijama. Stanice u njihovim organima, uključujući srce, jetru, bubrege i mozak, opet su bile u funkciji, a životinje ni u jednom trenutku nisu bile ukočene, kao što bi to bio slučaj s pravom mrtvom svinjom.

Ostale svinje, koje su bile mrtve već sat vremena, priključili su na umjetna pluća (ECMO uređaj), aparat koji je pumpao krv u njihova tijela. Ukočile su se, organi su im natekli i pretrpjeli oštećenja, a krvne žile su kolabirale. Na leđima su imale ljubičaste mrlje uslijed nakupljanja krvi.

Cilj - organi za transplantaciju

Skupina znanstvenika rezultate eksperimentalnog istraživanja objavila je u časopisu Nature.

Tvrde da je cilj njihova istraživanja povećati zalihe organa za transplantaciju. Nadaju se da bi se njihova tehnologija mogla koristiti za sprečavanje teških oštećenja srca - nakon razornog srčanog udara - ili mozga nakon moždanog udara.

No, najnovija otkrića tek su prvi korak, a još je daleko mogućnost da bi se slična tehnologija mogla isprobati na ljudima, rekao je Stephen Latham, bioetičar sa Sveučilišta Yale, koji je blisko surađivao sa znanstvenicima.

Skupinu znanstvenika predvodio je ugledni hrvatski znanstvenik Nenad Šestan, profesor neuroznanosti, komparativne medicine, genetike i psihijatrije na Medicinskom fakultetu Yaleu

i on je bio zapanjen otkrićem. Posjetimo, prije tri godine, osvanuo je na naslovnicama vodećih svjetskih medija zahvaljujući sličnom, velikom otkriću. Šestan i njegovi suradnici uspjeli su uz pomoć visokotehnološkog sustava BrainEx, koji su sami razvili, obnoviti temeljnu staničnu aktivnost u svinjskom mozgu nekoliko sati nakon smrti životinje. Popularnim jezikom rečeno, uspjeli su "oživjeti" stanice mozga jednog sisavca četiri sata nakon smrti.

Jedan od autora studije je i dr. David Andrijević, mladi Osječanin, također neuroznanstvenik s Yalea, prenosi Jutarnji list. 

- Nismo znali što možemo očekivati. Bilo je nevjerojatno, rekao je.

Nova definicija smrti?

I drugi znanstvenici koji nisu sudjelovali u studiji bili su podjednako zadivljeni.

- Nevjerojatno, zapanjeni smo, rekla je Nita Farahany, profesorica prava na Dukeu koja poučava etičke, pravne i društvene implikacije novih tehnologija.

Dr. Farahany je rekla da rad postavlja pitanja o definiciji smrti.

Rad na projektu započeo je prije nekoliko godina kada je skupina napravila sličan eksperiment sa svinjskim mozgom.

Prema riječima još jednog člana tima iz Hrvatske, dr. Zvonimira Vrselje, otkriće iz 2019. ih je navelo da se zapitaju mogu li oživjeti cijelo tijelo.

Otopina OrganEx sadrži hranjive tvari, protuupalne lijekove, lijekove za sprečavanje smrti stanica, blokatore živaca - tvari koje prigušuju aktivnost neurona i sprečavaju svaku mogućnost da svinje dođu svijesti - te umjetni hemoglobin pomiješan s krvlju svake životinje.

Znanstvenici su poduzeli sve mjere opreza kako životinje ne bi patile. Svinje su dobile anesteziju prije nego što im je zaustavljen rad srca, a duboka anestezija je nastavljena tijekom postupka.

Osim toga, blokatori živaca u otopini OrganEx zaustavljaju oštećenje živaca kako bi se osiguralo da mozak ne bude aktivan. Znanstvenici su također hladili životinje kako bi usporili kemijske reakcije. Pojedinačne moždane stanice bile su žive, ali nije bilo naznaka bilo kakve organizirane živčane aktivnosti u mozgu.

Jedan detalj bio je osobito zapanjujući: svinje koje su liječene OrganExom trzale su glavama kada su istraživači ubrizgali kontrastno sredstvo za snimanje.

Dr. Latham naglašava da ne znaju zbog čega su se svinje trzale, jer nije bilo naznaka da je mozak potaknuo kretnje.

Yale je podnio zahtjev za patentiranjem nove tehnologije. Prema riječima Nenada Šestana, njihov cilj je vidjeti funkcioniraju li organi kako treba i mogu li se uspješno presaditi.

Kroz određeno vrijeme, znanstvenici se nadaju da će testirati može li tehnologija pridonijeti oporavku oštećenog srca ili mozga.
