Veliko novo istraživanje bolesti Alzheimer znanstvenici su uspjeli otkriti protein u mozgu koji utječe na razvoj bolesti čime se otvara mogućnost da se njegovom daljnjom analizom stvore strategije kako ga regulirati i time naći načine kako pomoći oboljelima, ili čak spriječiti samu bolest, piše New Atlas.

Znanstvenici s Medicinskog fakulteta Sveučilišta Washington (WUSM) u St. Louisu proveli su nekoliko godina istražujući veze između biološkog dnevnog ritma i Alzheimerove bolesti, a nedavno se dogodio prvi pravi pomak. Nakon studije iz 2018. koja je pokazala kako poremećaj sna može ubrzati nakupljanje toksičnih naslaga povezanih s bolešću, tim je sada identificirao protein koji je dijelom odgovoran za ubrzanje bolesti. Čini se da na taj protein uvelike utječe cirkadijanski ritam koji označava psihološke i fiziološke promjene unutar perioda od 24 sata te izmjenu dana i noći u okolišu pojedinca.

U svom prethodnom istraživanju, tim WUSM-a krenuo je istraživati kako poremećaji naših prirodnih ciklusa spavanja ili cirkadijalnog ritma mogu ubrzati nakupljanje amiloidnih naslaga u mozgu, koje su čvrsto povezane s Alzheimerovom bolešću. Kroz studije na ljudima i na miševima, tim je uspio dokazati snažnu korelaciju između toga dvoje, a sada je kroz daljnji rad identificirao moždani protein koji, čini se, igra ulogu u ovom odnosu.

Sporni protein u mozgu naziva se YKL-40 i već godinama služi kao biomarker za Alzheimerovu bolest, jer su pronađene njegove visoke razine u likvoru oboljelih od te bolesti i njegova količina raste kako bolest napreduje. Istraživači su pregledavali gene koji su regulirani biološkim dnevnim ritmom i bili su zaintrigirani kad su vidjeli gen za ovaj protein u mozgu.

Odatle je tim istraživao vezu između YKL-40 i Alzheimerove bolesti, koju karakterizira kronična upala, istražujući koliko proteina nastaje u upalnim uvjetima uz prisutnost i bez ključnog cirkadijskog gena. To je doista pokazalo da cirkadijanski ritam kontrolira koliko se proizvodi YKL-40.

Voditelj znanstvenog istraživanja dr. Erik Museik objašnjava da se razlikuje količina YKL-40 ovisno o tome kada se dobije upala. Tako će se ujutro dobiti puno više proteina u slučaju upale, nego li ako se ona dogodi uvečer.

Tim je također radio s miševima sklonima razvoju amiloidnih naslaga i genetski modificirao jednu skupinu kako bi im nedostajao gen za YKL-40. Kako su miševi stasali, tim je analizirao njihov mozak i otkrio da oni bez proteina YKL-40 pokazuju upola manje amiloidnih naslaga od kontrolne skupine, tj. one kod koja nije genetski modificirana.

Pokušavajući dalje shvatiti uzroke te pojave, tim je otkrio da su miševi kojima nedostaje gen YKL-40 posjedovali više mikroglija, tj. imunoloških stanica koje okružuju amiloidne naslage i sprječavaju njihovo širenje. U osnovi je to značilo da su ti miševi imali više 'gladnih' imunoloških stanica pripremljenih za proždiranje amiloida.

-Ovaj protein YKL-40 vjerojatno služi kao regulator razine aktivacije mikroglija u mozgu. Kad se riješite proteina, čini se da se više mikroglija aktivira da pojedu amiloid. To je suptilna stvar, dotjerivanje sustava, ali čini se da je to dovoljno za značajno smanjenje ukupnog amiloidnog tereta, pojasnio je dr.Museik.

Tim je nakon toga ispitivao valjanost ove tvrdnje kod ljudi, oslanjajući se na genetske podatke o 778 ispitanika iz studija starenja i demencije. Otkrili su da je samo četvrtina njih imala genetsku varijantu koja snižava razinu YKL-40, a da je kognitivna funkcija sporije opadala za 16 posto u toj skupini.

-Ako vaš dnevni bioritam godinama nije sasvim u redu, tj. rutinski patite noću od nesanice i drijemate danju, kumulativni učinak kronične disregulacije mogao bi utjecati na upalne putove tako da nakupljate više amiloidnih naslaga. Nadamo se da bi bolje razumijevanje utjecaja cirkadijalnog sata na YKL-40 moglo dovesti do nove strategije za smanjenje amiloida u mozgu, završio je dr. Museik.

Drugim riječima otkrije li se kako smanjiti amiloidne naslage moglo bi se početi rješavati problem Alzheimera. Za sad se čini da bi se to moglo učiniti smanjenjem proteina YKL-40, a na njega utječe kvaliteta sna, te uravnoteženi bioritam.

Veliko novo istraživanje bolesti Alzheimer znanstvenici su uspjeli otkriti protein u mozgu koji utječe na razvoj bolesti čime se otvara mogućnost da se njegovom daljnjom analizom stvore strategije kako ga regulirati i time naći načine kako pomoći oboljelima, ili čak spriječiti samu bolest, piše New Atlas.

Znanstvenici s Medicinskog fakulteta Sveučilišta Washington (WUSM) u St. Louisu proveli su nekoliko godina istražujući veze između biološkog dnevnog ritma i Alzheimerove bolesti, a nedavno se dogodio prvi pravi pomak. Nakon studije iz 2018. koja je pokazala kako poremećaj sna može ubrzati nakupljanje toksičnih naslaga povezanih s bolešću, tim je sada identificirao protein koji je dijelom odgovoran za ubrzanje bolesti. Čini se da na taj protein uvelike utječe cirkadijanski ritam koji označava psihološke i fiziološke promjene unutar perioda od 24 sata te izmjenu dana i noći u okolišu pojedinca.

U svom prethodnom istraživanju, tim WUSM-a krenuo je istraživati kako poremećaji naših prirodnih ciklusa spavanja ili cirkadijalnog ritma mogu ubrzati nakupljanje amiloidnih naslaga u mozgu, koje su čvrsto povezane s Alzheimerovom bolešću. Kroz studije na ljudima i na miševima, tim je uspio dokazati snažnu korelaciju između toga dvoje, a sada je kroz daljnji rad identificirao moždani protein koji, čini se, igra ulogu u ovom odnosu.

Sporni protein u mozgu naziva se YKL-40 i već godinama služi kao biomarker za Alzheimerovu bolest, jer su pronađene njegove visoke razine u likvoru oboljelih od te bolesti i njegova količina raste kako bolest napreduje. Istraživači su pregledavali gene koji su regulirani biološkim dnevnim ritmom i bili su zaintrigirani kad su vidjeli gen za ovaj protein u mozgu.

Odatle je tim istraživao vezu između YKL-40 i Alzheimerove bolesti, koju karakterizira kronična upala, istražujući koliko proteina nastaje u upalnim uvjetima uz prisutnost i bez ključnog cirkadijskog gena. To je doista pokazalo da cirkadijanski ritam kontrolira koliko se proizvodi YKL-40.

Voditelj znanstvenog istraživanja dr. Erik Museik objašnjava da se razlikuje količina YKL-40 ovisno o tome kada se dobije upala. Tako će se ujutro dobiti puno više proteina u slučaju upale, nego li ako se ona dogodi uvečer.

Tim je također radio s miševima sklonima razvoju amiloidnih naslaga i genetski modificirao jednu skupinu kako bi im nedostajao gen za YKL-40. Kako su miševi stasali, tim je analizirao njihov mozak i otkrio da oni bez proteina YKL-40 pokazuju upola manje amiloidnih naslaga od kontrolne skupine, tj. one kod koja nije genetski modificirana.

Pokušavajući dalje shvatiti uzroke te pojave, tim je otkrio da su miševi kojima nedostaje gen YKL-40 posjedovali više mikroglija, tj. imunoloških stanica koje okružuju amiloidne naslage i sprječavaju njihovo širenje. U osnovi je to značilo da su ti miševi imali više 'gladnih' imunoloških stanica pripremljenih za proždiranje amiloida.

-Ovaj protein YKL-40 vjerojatno služi kao regulator razine aktivacije mikroglija u mozgu. Kad se riješite proteina, čini se da se više mikroglija aktivira da pojedu amiloid. To je suptilna stvar, dotjerivanje sustava, ali čini se da je to dovoljno za značajno smanjenje ukupnog amiloidnog tereta, pojasnio je dr.Museik.

Tim je nakon toga ispitivao valjanost ove tvrdnje kod ljudi, oslanjajući se na genetske podatke o 778 ispitanika iz studija starenja i demencije. Otkrili su da je samo četvrtina njih imala genetsku varijantu koja snižava razinu YKL-40, a da je kognitivna funkcija sporije opadala za 16 posto u toj skupini.

-Ako vaš dnevni bioritam godinama nije sasvim u redu, tj. rutinski patite noću od nesanice i drijemate danju, kumulativni učinak kronične disregulacije mogao bi utjecati na upalne putove tako da nakupljate više amiloidnih naslaga. Nadamo se da bi bolje razumijevanje utjecaja cirkadijalnog sata na YKL-40 moglo dovesti do nove strategije za smanjenje amiloida u mozgu, završio je dr. Museik.

Drugim riječima otkrije li se kako smanjiti amiloidne naslage moglo bi se početi rješavati problem Alzheimera. Za sad se čini da bi se to moglo učiniti smanjenjem proteina YKL-40, a na njega utječe kvaliteta sna, te uravnoteženi bioritam.

Veliki napredak u istraživanju opake bolesti: poremećaji u bioritmu utječu na razvoj proteina u mozgu kojeg se koristi kao biomarker za Alzheimera

Veliko novo istraživanje bolesti Alzheimer znanstvenici su uspjeli otkriti protein u mozgu koji utječe na razvoj bolesti čime se otvara mogućnost da se njegovom daljnjom analizom stvore strategije kako ga regulirati i time naći načine kako pomoći oboljelima, ili čak spriječiti samu bolest, piše New Atlas.

Znanstvenici s Medicinskog fakulteta Sveučilišta Washington (WUSM) u St. Louisu proveli su nekoliko godina istražujući veze između biološkog dnevnog ritma i Alzheimerove bolesti, a nedavno se dogodio prvi pravi pomak. Nakon studije iz 2018. koja je pokazala kako poremećaj sna može ubrzati nakupljanje toksičnih naslaga povezanih s bolešću, tim je sada identificirao protein koji je dijelom odgovoran za ubrzanje bolesti.
